Vitamin K dependent anticoagulants

Abstract
Warfarin is an anticoagulant that inhibits the synthesis of several vitamin K-dependent coagulation factors. Therefore, it is widely used to prevent thrombosis and thromboembolism in patients with atrial fibrillation, mechanical valves, or deep vein thrombosis. Despite its efficacy, warfarin has multiple drawbacks, which makes clinical administration difficult and increases the risk of undesirable side effects. Warfarin has a narrow therapeutic window, with supratherapeutic levels carrying the risk of hemorrhage and subtherapeutic levels predisposing toward systemic thromboembolism. Environmental modulation from diet and concurrent drug administration are also major problems. An extensive understanding of warfarin is needed for its effective clinical application.
